QRICH1 is a negative regulator of T cell activation via the CARD11 signalosome

چکیده

Abstract Controlled lymphocyte activation is crucial for defending against pathogens while preventing diseases such as autoimmunity and lymphoma. In T lymphocytes, the scaffold protein signaling hub CARD11 mediates NF-κB, JNK, mTOR pathway downstream of cell receptor (TCR) engagement. We recently identified QRICH1 a previously unrecognized interactor recruited to complex TCR engagement discovered that levels are upregulated in wild-type activated mouse primary CD8+ cells. Overexpression 293T Jurkat cells dose-dependently inhibited NF-κB by well constitutively active oncogenic mutants. QRICH1-deficient had increased IL-2 secretion stimulation. To study vivo role immunity, we generated novel conditional KO model. Compared control mice, mice decreased splenic CD4+ cells, despite no differences thymic CD4+, CD8+, or CD4+CD8+ populations. The population was skewed towards an effector memory phenotype. proliferation production pro-inflammatory cytokines IFN-γ TNF-α response ex stimulation with anti-CD3/CD28, OT-1 also cytokine antigen-specific Ova peptide. Our findings reveal negatively regulates modulating CARD11’s output. This work supported NIH grants R01AI148143, F31CA254167, T32GM007445.

DC-HIL is a negative regulator of T lymphocyte activation.

T-cell activation is the net product of competing positive and negative signals transduced by regulatory molecules on antigen-presenting cells (APCs) binding to corresponding ligands on T cells. Having previously identified DC-HIL as a receptor expressed by APCs that contains an extracellular immunoglobulin (Ig)-like domain, we postulated that it plays a role in T-cell activation. To probe this...

Dok-4 is a novel negative regulator of T cell activation.

Dok-4 (downstream of tyrosine kinase-4) is a recently identified member of the Dok family of adaptor proteins, which are characterized by an amino-terminal pleckstrin homology domain, a phosphotyrosine-binding domain, and a carboxyl-terminal region containing several tyrosines and poly-proline-rich motifs. Two members of the Dok family, Dok-1 and Dok-2, have already been described as negative r...

Perforin is a critical physiologic regulator of T-cell activation.

Individuals with impaired perforin-dependent cytotoxic function (Ctx(-)) develop a fatal inflammatory disorder called hemophagocytic lymphohistiocytosis (HLH). It has been hypothesized that immune hyperactivation during HLH is caused by heightened infection, defective apoptosis/responsiveness of Ctx(-) lymphocytes, or enhanced antigen presentation. Whereas clinical and experimental data suggest...

B and T Lymphocyte Attenuator is a Target of miR-155 during Naive CD4+ T Cell Activation

Background: MicroRNA-155 (miR-155) is upregulated during T cell activation, but the exact mechanisms by which it influences CD4+ T cell activation remain unclear. Objective: To examine whether the B and T lymphocyte attenuator (BTLA) is a target of miR-155 during naïve CD4+ T cell activation. Methods: Firefly luciferase reporter plasmids pEZX-MT01-wild-type-BTLA and pEZX-MT01-mutant-BTLA were ...
